Gillingham will play Colchester United, Wycombe Wanderers and Fulham U21s in the group stage of the 2025/26 Vertu Trophy.
The clubs have been placed in Group G of the southern section with opening games set to be played in September.
The Final will take place at Wembley Stadium on Sunday April 12.
